Description

The freshly-harvested animal tissue is flash-frozen in liquid nitrogen in less than 30 seconds upon removal and stored via submersion in liquid nitrogen until cryoground to a fine powder using a mortar and pestle at less than -80°C. This powder is thoroughly homogenized at 5°C with a freshly prepared modified, inhibited Radio Immuno Precipitation Assay buffer containing strong protease and phosphatase inhibitors. The final homogenate is clarified by centrifugation at 10,000 g (0°C) and stored at -80°C; samples are shipped on dry ice. Each lot is quality controlled by determining the protein concentration using a BCA assay and analyzed with SDS-PAGE using Coomassie blue for visualization. Sharp, clear protein bands indicate a quality protein lysate.
